    Ground effect for V/STOL aircraft configurations and its simulation in the wind tunnel. Part 1: Introduction and theoretical studies

    Theoretical studies are made of three dimensional turbulent boundary layer behavior on fixed grounds and on moving grounds of the type used in wind tunnel tests. It is shown that, for several widely-varying STOL configurations, the ground static pressure distributions possess a remarkable degree of fore-aft symmetry about the center of lift. At low Renolds number, corresponding to small-tunnel testing, the boundary layer displacement surface reflects to a large degree the symmetry of the pressure distribution. For this reason, induced incidence at the model is small for unseparated ground flow. At high Reynolds number, the displacement thickness decrease aft of the static pressure maximum is noticeably more rapid than the corresponding rise. This is attributed to trailing-vortex-induced spanwise pumping within the boundary layer

    A longitudinal study of the personal and professional responses of recently assigned secondary Religious Education teachers to curriculum demands

    This research is a longitudinal study, conducted in Catholic schools of Perth and its environs. From 1998 to 1999, recently appointed secondary Religious Education teachers were surveyed and interviewed, and re-interviewed eighteen months later. Initially, a comprehensive survey of three dioceses in Western Australia was conducted and formed a contextual database for the study. The database acted as the foundation for the selection of a purposive sample of teachers for the first round of interviews. Follow-up interviews with this sample cohort were carried out the following year. The research design used in this study assisted the development of a representative, grounded analysis of the perceptions of recently assigned RE teachers. In the first interview, participants were asked to comment on their experiences of implementing a new draft RE program in Catholic schools in Western Australia. The teachers were invited to relate their experiences and perceptions of implementing the curriculum materials, the advised teaching approaches and understandings of the theological and pedagogical principles underlying the new Units. In the second interview, teachers were invited to reflect upon their personal and professional growth as they became more familiar with the demands of this RE curriculum. What emerged were insights into the nature and depth of formation these teachers experienced as they introduced the draft Units of Work. Using qualitative analysis techniques such as NUD•IST, findings emerged about the importance of the personal spiritual and faith formation of teachers during this period. Teachers felt passionate about why they were teaching RE and implemented the Units with enthusiasm. They were initially optimistic about the future of their RE teaching but were then confronted with challenges to their personal, spiritual and faith formation. Most teachers continued to look forward to teaching RE, while some were relieved when they had the opportunity to discontinue. These findings suggest that there is a need to consider how these teachers can be personally and professionally supported as they face the transition from a ‘crusade of delivery’ to a ‘pilgrimage of formation’ in their RE teaching lives

    If you have taught Religious Education in a Catholic secondary school, then you may have sat down at the end of the class or the school day and just wondered why you bothered. Before reading this article further, give yourself a few moments to reflect on this question: “What for you is the best and most special about teaching Religious Education?” How did you answer the question? Was your response negative or positive? Did you reach for the instant cynical remark, “When I finished teaching the class!” Or, did you imagine yourself taking great pride in accomplishing deeper understandings among your students? Maybe your response was one of mixed feelings. These responses are important because the disposition of a teacher towards his or her RE teaching is a key factor in the quality of Religious Education provided to secondary students in Catholic schools. This article outlines the findings of the second phase of a study about the teaching experiences of recently assigned RE teachers and the resultant changes in their outlook about RE teaching

    Changes in yield and composition of barley, wheat and triticale grains harvested during advancing stages of ripening

    peer-reviewedThis study involved an evaluation of the changes in grain yield, nutritive value, ensilability and harvesting losses of intensively managed winter cereals harvested during the advancing stages of ripening. Five cereal crops (barley cv. Regina and wheat cv. Madrigal in 2001; barley cv. Regina, wheat cv. Falstaff and triticale cv. Fidelio in 2002) were assessed. Twenty plots per crop were arranged in a randomised complete block design, with five times of harvest (four for barley in 2002) and four replicate blocks per harvest. Dry matter (DM) yields changed relatively little between harvest dates, but fresh yields declined (P < 0.001) over time due to the moisture loss associated with ripening. Time-course changes in indices of nutritive value, such as concentrations of crude protein, starch and ash, and organic matter digestibility, were relatively small and did not follow a consistent pattern. Ensilability indices, such as DM and watersoluble carbohydrate concentrations and buffering capacity, indicated that satisfactory fermentations were likely if such crops were ensiled; buffering capacity, generally declining with advancing maturity. Harvesting losses were not clearly related to growth stage at harvest. It is concluded that winter cereal grain (barley, wheat and triticale) DM yields and quality were relatively constant as ripening progressed from DM concentrations of around 550 to >800 g/kg.One of the authors (P.S.) was in receipt of a Teagasc Walsh Fellowship

    Predicting preferential DNA vector insertion sites: implications for functional genomics and gene therapy

    Viral and transposon vectors have been employed in gene therapy as well as functional genomics studies. However, the goals of gene therapy and functional genomics are entirely different; gene therapists hope to avoid altering endogenous gene expression (especially the activation of oncogenes), whereas geneticists do want to alter expression of chromosomal genes. The odds of either outcome depend on a vector's preference to integrate into genes or control regions, and these preferences vary between vectors. Here we discuss the relative strengths of DNA vectors over viral vectors, and review methods to overcome barriers to delivery inherent to DNA vectors. We also review the tendencies of several classes of retroviral and transposon vectors to target DNA sequences, genes, and genetic elements with respect to the balance between insertion preferences and oncogenic selection. Theoretically, knowing the variables that affect integration for various vectors will allow researchers to choose the vector with the most utility for their specific purposes. The three principle benefits from elucidating factors that affect preferences in integration are as follows: in gene therapy, it allows assessment of the overall risks for activating an oncogene or inactivating a tumor suppressor gene that could lead to severe adverse effects years after treatment; in genomic studies, it allows one to discern random from selected integration events; and in gene therapy as well as functional genomics, it facilitates design of vectors that are better targeted to specific sequences, which would be a significant advance in the art of transgenesis

    First-year impact of an Early Reading First project on language and early reading skill development

    This study is of a federally-funded Early Reading First project aimed at improving school readiness by creating Centers of Excellence which focus on language and early reading skills development of low-income preschool children. The project consisted of a content standards-based curriculum and instructional program, literacy enriched classrooms, ongoing professional development, and parent involvement. The analysis focuses on 65 randomly assigned preschoolers attending state preschools (treatment = 37 and control = 28), enrolled for at least 18 weeks, who would be eligible by birth date for entry into public kindergarten the following year. Results demonstrate the intervention to be effective in improving the receptive language and early reading skills of participating preschoolers

    Insights into a dinoflagellate genome through expressed sequence tag analysis

    BACKGROUND: Dinoflagellates are important marine primary producers and grazers and cause toxic "red tides". These taxa are characterized by many unique features such as immense genomes, the absence of nucleosomes, and photosynthetic organelles (plastids) that have been gained and lost multiple times. We generated EST sequences from non-normalized and normalized cDNA libraries from a culture of the toxic species Alexandrium tamarense to elucidate dinoflagellate evolution. Previous analyses of these data have clarified plastid origin and here we study the gene content, annotate the ESTs, and analyze the genes that are putatively involved in DNA packaging. RESULTS: Approximately 20% of the 6,723 unique (11,171 total 3'-reads) ESTs data could be annotated using Blast searches against GenBank. Several putative dinoflagellate-specific mRNAs were identified, including one novel plastid protein. Dinoflagellate genes, similar to other eukaryotes, have a high GC-content that is reflected in the amino acid codon usage. Highly represented transcripts include histone-like (HLP) and luciferin binding proteins and several genes occur in families that encode nearly identical proteins. We also identified rare transcripts encoding a predicted protein highly similar to histone H2A.X. We speculate this histone may be retained for its role in DNA double-strand break repair. CONCLUSION: This is the most extensive collection to date of ESTs from a toxic dinoflagellate. These data will be instrumental to future research to understand the unique and complex cell biology of these organisms and for potentially identifying the genes involved in toxin production

    Group cognitive behavioural therapy for stroke survivors with depression and their carers

    Background: Depression in stroke survivors is common, leads to poorer outcomes and often not treated. A group cognitive behavioural therapy (CBT) program (Brainstorm) for stroke survivors with depression, and their carers has been running as part of usual care since 2007. Objective: To evaluate the implementation and acceptability of Brainstorm, a closed group intervention consisting of up to 10 sessions of education, activity planning, problem solving and thought challenging. Methods: Participating stroke survivors and their carers complete assessment measures at baseline, post-treatment and 1-month and 6-months follow-up. A mixed models for repeated measures data was conducted with depression and anxiety scores for stroke survivors (Beck Depression Inventory-II; Hospital Anxiety and Depression Scale) and the assessment of depression, anxiety and carer burden for carers. Acceptability was assessed by session attendance and written and open participant feedback upon completion of the program. Results: Forty-eight community dwelling stroke survivors and 34 carers attended Brainstorm, with a median attendance of 88% of sessions. Follow-up assessments were completed by 77% (post-treatment), 46% (1-month) and 38% (6-month) of stroke survivors. Stroke survivors’ depression scores decreased from baseline to post-treatment (p<.001); maintained at 1-month (p<.001) but not at 6-month (p=.056). Anxiety scores decreased between baseline and 1-month (p=.013). Carer burden, depression and anxiety scores at 1-month and 6-month follow-up, for carers, were all reduced when compared with baseline (p<.05). Conclusion: The Brainstorm group intervention for depression in stroke survivors appears to have been effectively implemented and is acceptable to stroke survivors and carers

    Excision of Sleeping Beauty transposons: parameters and applications to gene therapy

    A major problem in gene therapy is the determination of the rates at which gene transfer has occurred. Our work has focused on applications of the Sleeping Beauty (SB) transposon system as a non-viral vector for gene therapy. Excision of a transposon from a donor molecule and its integration into a cellular chromosome are catalyzed by SB transposase. In this study, we used a plasmid-based excision assay to study the excision step of transposition. We used the excision assay to evaluate the importance of various sequences that border the sites of excision inside and outside the transposon in order to determine the most active sequences for transposition from a donor plasmid. These findings together with our previous results in transposase binding to the terminal repeats suggest that the sequences in the transposon-junction of SB are involved in steps subsequent to DNA binding but before excision, and that they may have a role in transposase-transposon interaction. We found that SB transposons leave characteristically different footprints at excision sites in different cell types, suggesting that alternative repair machineries operate in concert with transposition. Most importantly, we found that the rates of excision correlate with the rates of transposition. We used this finding to assess transposition in livers of mice that were injected with the SB transposon and transposase. The excision assay appears to be a relatively quick and easy method to optimize protocols for delivery of genes in SB transposons to mammalian chromosomes in living animals.
